The Animal Models for Dengue Virus Infection: Chapter 20
نویسندگان
چکیده
Currently, the mechanisms involved in the pathogenesis of DHF/DSS remain poorly understood and there is no effective vaccine available to prevent infection with DEN virus. The lack of a reliable small animal model that mimics dengue disease is a major obstacle. In this paper, the development of small animal models such as mice for dengue virus infections is reviewed.

Animal Models of Dengue Virus Infection
The development of animal models of dengue virus (DENV) infection and disease has been challenging, as epidemic DENV does not naturally infect non-human species. Non-human primates (NHPs) can sustain viral replication in relevant cell types and develop a robust immune response, but they do not develop overt disease. In contrast, certain immunodeficient mouse models infected with mouse-adapted D...
متن کاملA Model of DENV-3 Infection That Recapitulates Severe Disease and Highlights the Importance of IFN-γ in Host Resistance to Infection
There are few animal models of dengue infection, especially in immunocompetent mice. Here, we describe alterations found in adult immunocompetent mice inoculated with an adapted Dengue virus (DENV-3) strain. Infection of mice with the adapted DENV-3 caused inoculum-dependent lethality that was preceded by several hematological and biochemical changes and increased virus dissemination, features ...
متن کاملThe Intestinal Zonulin and Zonula Occludens 1 Protein Expression and Lipopolysaccharide Levels In ddY Mice Injected with Dengue Virus Non-Structural Protein 1
Background and Objective: Dengue virus infection remains a health problem. Dengue Virus Non-Structural protein 1 (NS1) increases the release of proinflammatory cytokines that induce intestinal zonulin expression. As a result, the ZO-1 protein translocates to the cytoplasm, which increases enterocyte permeability. This study aimed to investigate the effects of dengue NS1 on intestinal zonulin an...
متن کاملNeutralization of antibody-enhanced dengue infection by VIS513, a pan serotype reactive monoclonal antibody targeting domain III of the dengue E protein
Dengue virus (DENV) infection imposes enormous health and economic burden worldwide with no approved treatment. Several small molecules, including lovastatin, celgosivir, balapiravir and chloroquine have been tested for potential anti-dengue activity in clinical trials; none of these have demonstrated a protective effect. Recently, based on identification and characterization of cross-serotype ...
متن کاملDengue Virus Tropism in Humanized Mice Recapitulates Human Dengue Fever
Animal models of dengue virus disease have been very difficult to develop because of the virus' specificity for infection and replication in certain human cells. We developed a model of dengue fever in immunodeficient mice transplanted with human stem cells from umbilical cord blood. These mice show measurable signs of dengue disease as in humans (fever, viremia, erythema and thrombocytopenia),...
